Discovery and characterization of an endogenous CXCR4 antagonist.

چکیده
CXCL12-CXCR4 signaling controls multiple physiological processes and its dysregulation is associated with cancers and inflammatory diseases. To discover as-yet-unknown endogenous ligands of CXCR4, we screened a blood-derived peptide library for inhibitors of CXCR4-tropic HIV-1 strains. This approach identified a 16 amino acid fragment of serum albumin as an effective and highly specific CXCR4 antagonist. The endogenous peptide, termed EPI-X4, is evolutionarily conserved and generated from the highly abundant albumin precursor by pH-regulated proteases. EPI-X4 forms an unusual lasso-like structure and antagonizes CXCL12-induced tumor cell migration, mobilizes stem cells, and suppresses inflammatory responses in mice. Furthermore, the peptide is abundant in the urine of patients with inflammatory kidney diseases and may serve as a biomarker. Our results identify EPI-X4 as a key regulator of CXCR4 signaling and introduce proteolysis of an abundant precursor protein as an alternative concept for chemokine receptor regulation.

The chemokine receptor CXCR4 is an important G protein-coupled receptor. Signaling via CXCL12 regulates a number of important biologic processes, including immune responses, organogenesis, or hematopoiesis. Dysregulation of CXCR4 signaling is associated with a variety of diseases, such as cancer development and metastasis, immunodeficiencies, or chronic inflammation. Here, we review our finding...
متن کاملEPI-X4, a novel endogenous antagonist of CXCR4
Activation of CXC-motif-chemokine receptor 4 (CXCR4) signaling by the chemokine CXCL12 (also known as SDF-1α) plays a key role in many physiological processes including organogenesis, hematopoiesis, and immune responses. Consequently, CXCR4 or CXCL12 knockouts in mice are embryonic lethal because of impaired hematopoiesis, defects in organ development, and vascularization. Deregulation of CXCR4...

In light of a proposed molecular mechanism for the C-X-C chemokine receptor type 4 (CXCR4) antagonist 1 (AMD3100), a template with the general structure 2 was designed, and 15 was identified as a lead by means of an affinity binding assay against the ligand-mimicking CXCR4 antagonist 3 (TN14003). Following a structure-activity profile around 15, the design and synthesis of a series of novel sma...

The chemokine CXCL12 (SDF-1) and its cognate receptor CXCR4 are involved in a large number of physiological processes including HIV-1 infectivity, inflammation, tumorigenesis, stem cell migration, and autoimmune diseases. While previous efforts have identified a number of CXCR4 antagonists, there have been no small molecule agonists reported. Herein, we describe the identification of a novel se...

The present study investigated the effects of intra-dentate gyrus microinjection of naloxone (an opioid antagonist) and thioperamide (an antagonist of histamine H3 receptors) in the formalin test in rats. Subcutaneous injection of formalin (50 μl, 2.5 %) in the ventral surface of right hind paw produced a biphasic pattern (first phase: 0-5 min and second phase: 15 - 60 min) of licking/biting an...
